Molecular Characterization, Optimization and Production of PHA by Indigenous Bacteria Using Alternative Nutrient Sources as Substrate

Polyhydroxyalkanoates (PHA) are renewable, biodegradable biopolymer intracellularly accumulated by wide array of microorganisms as carbon reserve. This study investigates the influence various cultural conditions on PHA production a recently isolated local species under submerged fermentation. Six producing strains were identified 16S rDNA gene sequencing and strain Priestia flexa OWO1 showed satisfactory productivity. The effects parameters investigated extraction was carried out using sodium hypochlorite method maximum amount detected after 72h. Maximum obtained at incubation period 48h, pH 7.0 temperature 30oC. Amongst hydrolysate agro waste used, brewers spent grain (BSG) gave yield 3.01g/L while beans bran powder highest yields 3.9 g/L amongst organic nitrogen sources tested. Analysis crude Fourier Transform Infrared Spectroscopy (FT-IR) presence methyl, methylene well carbonyl functional groups. higher optimizing compared to basal medium therefore utilization these cheap renewable resources alternative substrates for make process cost effective sustainable.
